The regiment saw action primarily in the Eastern Theater of the war, participating in significant battles such as the First Battle of Bull Run, Antietam, Fredericksburg, and Gettysburg. Notably, the 2d Rhode Island Infantry is remembered for its valor during the Battle of Fort Wagner on July 18, 1863, in which it suffered heavy casualties while attempting to storm the fortifications held by Confederate forces.
